I've had the same problem of being sometimes unable to delete AI files (Windows XP pro, currently IL CS but similar problems with previous editions episodically). Turning off the computer and turning it back on frees up whatever lock is being set. I run no virus software and taskmaster shows no other programs running. It is only an occasional nuisance for me, but it seems unique to Illustrator. (even Corel Draw doesn't do it).
