    Yes I have it there in page_load event but it doesn't
    work...

    make sure that you recreate the grid on post back :

    private void Page_Load(object sender, System.EventArgs e)
    {
    FillData();
    }
    private void FillData()
    {
    ButtonColumn bcColumn = new ButtonColumn();
    bcColumn.HeaderText = "SubItems";
    bcColumn.ItemStyle.HorizontalAlign = HorizontalAlign.Center;
    bcColumn.ButtonType = ButtonColumnType.PushButton;
    bcColumn.Text = "SHOW";
    bcColumn.CommandName = "SubItem";
    DataGrid1.Columns.Add(bcColumn);
