If you're on a system with relatively limited RAM, and you have a large item on your clipboard (copied) this can force Illustrator to spend considerable time writing data to your hard drive before completing an Open operation.
Could also be server traffic problems, if you're opening from a network.
Could be a lot of things, few of which have to do with Illustrator. You'd need to describe the process in more detail.

My entire system slows down if I have our server on my desktop. We have about 20 people who go to it. I suggest you copy what you need to off of it and get rid of it before working on anything.
